Nitric oxide is synthesized from L-arginine by nitric oxide synthase (NOS) enzymes located in vascular endothelial cells. The reaction converts L-arginine, oxygen, and NADPH into L-citrulline, nitric oxide, and NADP⁺, with NO then diffusing into surrounding smooth muscle cells to promote vasodilation.
